Company Overview:

Prime Medicine is a leading biotechnology company dedicated to creating and delivering the next generation of gene editing therapies to patients. The Company is leveraging its proprietary Prime Editing platform, a versatile, precise and efficient gene editing technology, to develop a new class of differentiated, one-time, potentially curative genetic therapies. Designed to make only the right edit at the right position within a gene while minimizing unwanted DNA modifications, Prime Editors have the potential to repair almost all types of genetic mutations and work in many different tissues, organs and cell types.

Prime Medicine is currently progressing a diversified portfolio of eighteen programs initially focused on genetic diseases with a fast, direct path to treating patients or with a high unmet need because they cannot be treated using other gene-editing approaches. Over time, the Company intends to maximize Prime Editing’s therapeutic potential and advance potentially curative therapeutic options to patients for a broad spectrum of diseases. Position Overview

We are seeking a motivated Scientist/Sr. Scientist with broad expertise in genome analysis and cellular phenotyping to join our off-target group and contribute to our understanding of Prime Editing outcomes in the context of correcting genetic diseases. In this position the candidate will work with the off-target team to develop and implement assays to evaluate genomic, chromosomal, and structural off- and on- target events associated with Prime Editing. The candidate will collaborate closely with disease programs teams and their work will inform selection of lead candidates for therapeutic programs. A successful candidate will have substantial NGS experience, exposure to gene editing, and a deep understanding of assay development. The ideal candidate will have advanced analytical skills, thrive in a fast-paced environment and be comfortable working in close collaboration with multiple teams including, disease biology, computational sciences and protein chemistry to execute the company’s off-target strategies.

Key Responsibilities:

  • Develop, implement, and validate assays (including NGS assays) to measure off- and on-target consequences of Prime Editing
  • Contribute Cellular phenotyping and NGS expertise as part of a team developing a suite of cell biological and genomic assays for comprehensive assessment of Prime Editing
  • Collaborate on cross-functional program teams to build and apply fit-for purpose methods that facilitate tiering of hits toward selection of development candidates
  • Deliver reproducible and impactful results under ambitious timelines
  • Adhere to all laboratory safety requirements and procedures, assist in maintaining general laboratory functionality
  • Author, review, and approve protocols, SOPs, technical documents, and reports in support of regulatory submissions

Qualifications

  • PhD with substantial academic and/or industry experience including a track record of impactful contributions. BSc/MSc with 5+ years of industry experience may also be considered.
  • Experience with molecular biology techniques, including NGS, DNA/RNA isolation and quantification, PCR, and sanger sequencing.
  • Experience with end-to-end NGS experiments including library preparation, sequencing, and analysis for at least one common modality (eg. mRNA-seq, ChIP-seq, ATAC-seq)
  • Familiarity with cell culture techniques including transfection, cell line generation, genomic DNA isolation, and RNA isolation.
  • Experience analyzing genomic structure in a gene editing context and/or long read sequencing is a plus
  • Experience with histological samples, biodistribution studies, or AAV analysis is a plus
  • Interest and background in computational analysis or data handling is a substantial plus
  • Demonstrated maturity to lead with influence, work independently, and cohesively as a self-aware team player
  • Strong oral, written, and verbal communication skills and the ability to work in a highly collaborative, fast-paced environment
